The announcement is accompanied by a trailer introducing assassin group 'The Saints', of which Fox plays head nun Lasandra Dixon.
The video explores the origins of 'The Saints', their methods and missions, including their latest job to eliminate Agent 47.
Square Enix has also released an interview with the actress herself, who talks about her role in the title and the similarities between making video games and shooting movies.
The Independence Day and Batman and Robin star describes her character as a "bad bitch", who isn't afraid to get her hands dirty on a job.
The first new Hitman game in six years, Hitman: Absolution will include new gameplay features, improved enemy intelligence and a new 'Instinct' mode that will help players blend into certain roles.
Described as the most ambitious game in the series yet, the stealth title features a "living and breathing world, where every moment can become a story".
IO Interactive told Digital Spy that Hitman: Absolution will have a "more personal story" than previous games.
Hitman: Absolution will be available on Xbox 360, PS3 and PC from November 20 worldwide.
Pre-order customers can also bag the Hitman Sniper Challenge mini-game, as well as the recently announced Hitman Absolution Deluxe Professional Edition.
