The actor - who portrays rebel slave Oenomaus in Starz's Spartacus miniseries - will appear in the recurring role of Kibwe, according to Deadline.
Kibwe is a chancellor for the Vampire Authority who hails from Africa, and will play a key role in mainstreaming under Roman's tenure.
Mensah has also appeared in TV series such as The Sarah Connor Chronicles and Star Trek: Enterprise.
His previous film roles include Avatar, 300 and Hidalgo.
Superman: Man of Steel star Christopher Meloni was recently confirmed as a series regular for season five of the show, with showrunner Alan Ball suggesting that Meloni's character will have control over the fate of Bill and Eric.
Season five of True Blood premieres later this year on HBO in the US, while season four is currently airing on Sundays at 10pm on FX in the UK.
